The aim of this proposal is to develop techniques for macroscopic imaging of whole-brain regions at a resolution that enables quantification of fine microscopic structural details. Development of this technology will enable alterations in neuronal connectivity to be studied both during the normal ageing process and following neurodegeneration. Mapping of neuronal projections will be undertaken in mouse models using novel viral delivery strategies. High-resolution imaging of this type is extremely data-intensive, and new analysis tools will be developed in order to process and analyse these complex data-sets.
